Activity

Pharmacological action

Antifungal, fungistatic. The fungistatic action is bounded with influence on microtubular proteins of fungal cells, it results in a breakage of fusiform mitotic structures and a stopping of mitotic division of fungal cells in metaphase, inhibits protein synthesis, breaks formation of cellular walls at chitin-containing fungi. It has more tropism to the affected tissues: it selectively accumulates in precursors of skin, hair and fingernail keratinocytes, strongly binds with neogenic keratin, providing high resistance to fungal invasion (thus it prevents mycotic affection of new formed cells). The keratin-containing structures are gradually exfoliated. Recovery comes after full changing of infected keratin therefore the medical effect develops slowly.
It is active against dermatomycetes, including Trichophyton (T. rubrum, T. tonsurans, T. mentagrophytes, T. interdigitalis, T. verrucosum, T. sulphureum, T. menginii, T. gallinae, T. schoenleini, T. crateriforme), Microsporum (M. audouinii, M. canis, M. gypseum), Epidermophyton (E. floccosum). Fungistatic concentration for the majority of fungi species sensitive to griseofulvin is 0,5–3 μg/ml. Yeast-like fungi (Candida genus) and mould fungi are resistant to griseofulvin. It doesn’t have antibacterial activity.

After oral administration it accumulates in corneous layer of skin and its appendages, and also in liver, fatty tissue, sceletal musculature. Insignificant quantities penetrate to fluid mediums and other tissues of an organism.

Mycoses of skin, scalp and fingernails: favus, trichophytosis, microsporia of scalp, microsporia of smooth skin, dermatomycosis of beard and moustaches, epidermophitia of smooth skin, inguinal epidermophitia, onychomycoses.
